β¦ Synopsis
A specific and sensitive electron-capture gas-liquid chromatographic method was devised for the estimation of piperidine, a putative neurotransmitter, in rat and dog brain tissue. The procedure involves the addition of an internal standard, 2-ethylpiperidine, to brain homogenates followed by extraction of the amines into benzene at an alkaline pH. The amines are back-extracted into acid and subsequently reextracted into benzene under alkaline conditions. The isolated amines are converted to the trichloroacetyl derivatives prior to chromatographic analysis. The sensitivity of the method is such that 3 ng of piperidine (35 pmol) per aliquot of brain homogenate can be analyzed. Combined gas-liquid chromatography/mass spectrometry confirmed the specificity of analysis. No significant difference was observed in the regional brain distribution of piperidine in the rat (-30 @pm) or the dog (-20 @pm).
