โฆ Synopsis
The characteristic red color produced when ninhydrin reacts with proline at pH 1 was first used by Chinard (1) for the estimation of this imino acid. Modifications of this method include removal of basic amino acids on Permutitl cation exchanger and extraction of the colored product into benzene (2), and destruction of other amino acids with nitrous acid (3) or performic acid (4). Messer (5) reported enhancement of the proline color and interferences by various amino acids, dipeptides, and tripeptides and partly overcame these interferences by adding glycine in excess to the ninhydrin reagent. This paper describes the use of the Technicon Auto-Analyzer for the automation of the Chinard method for the quantitative determination of proline.
