Complete plants have been obtained from 54 individually cultured haploid protoplasts. However, the technique can be used, if necessary, to regenerate thousands of individual cells into plants. No divisions have ever occurred in single-cell cultures when microdroplet volumes larger than 2 ~tl were used, regardless of the media employed. Thus, the experiments provide clear-cut demonstration of the fact that only diffusion of certain substances accounts for the failure of the plant cells to grow in diluted suspensions.

Thirdly, we have also found that preculturing of the protoplasts for 1 3 days in suspensions at high densities (104-105 cells/ ml) can significantly improve the plating efficiency of these cells in individual culture as illustrated in Table 1. Preculturing
