β¦ Synopsis
Perturbation of cellular thiol homeostasis occurs during the early phase of oxidative cell injury in isolated hepatocytes. This perturbation, involves depletion of soluble and protein-bound thiols, and is associated with altered intracellular calcium (Ca") homeostasis resulting from inhibition of major Ca2+ transport systems. The result is sustained increase in cytosolic Ca2+ concentration which appears to trigger cytotoxicity.
The step-wise reduction of molecular oxygen to water results in formation of several, potentially toxic, intermediates, including superoxide anion-free radical, hydrogen peroxide and the hydroxyl radical:
O2 + ee 0; 0; + e-+ 2H' + H202 H202 + e-+ H' e OH. + H20 OH. + e-+ H' + H20
