A serum-free culture system was established for human KB carcinoma (HeLa) cells that consisted of a chemically defined medium and several growth factors including epidermal growth factor (EGF), insulin, transferrin, hydrocortisone, and ethanolamine. EGF and insulin showed the greatest effects on the

โฆ Synopsis
Nostrand, McKinley, and Cunningham (unpublished results) have shown that one can resolve intracellular processed forms of EGF by native gel electrophoresis. Using this method to study the fate of internalized EGF in KB cells, we report herein the appearance of three intracellular intermediates in the degradation of 1251-EGF prior to the release of 1251-iodotyrosine into the medium and have attempted to determine the cellular locations at which these intermediates were produced. This process was altered in a cell line, KB-RBA, that is resistant to diphtheria toxin and to infection by RNA viruses.
Studies concerning the endocytic pathway are also relevant to the mechanism of action of diphtheria toxin (see Collier, 1975;and Pappenheimer, 1977; for reviews).
