✦ Synopsis


A l k a l o p h i l i c Bacillus sp. A G -4 3 0 was isolated from soil a n d f o u n d to p r o d u c e an extracellular fl-l,3-glucanase. This enzyme was purified by c h r o m at o g r a p h y on D E A E -s e p h a r o s e CL-6B, S e p h a d e x G-75 a n d h y d r o x y a p a t i t e . The enzyme was extremely therm o s t a b l e a n d lost only 10% of the original activity after i n c u b a t i o n at 100 ° C for 10 min. The o p t i m u m t e m p e r ature and p H for activity were 6 0 °-6 5 °C and 9-10, respectively. The m o l e c u l a r weight was estimated at a b o u t 35 000 on s o d i u m d o d e c y l s u l p h a t e -p o l y a c r y l a mide gel e l e t r o p h o r e s i s a n d the p I was a b o u t 3.8. The enzyme h y d r o l y s e d laminaritetraose, but not laminaritriose, a n d the e n d -p r o d u c t s detected in the h y d r o l y s a t e were identified as glucose, l a m i n a r i b i o s e a n d laminaritriose. The enzyme split l a m i n a r i n at r a n d o m a n d y i e l d e d glucose, l a m i n a r i b i o s e , laminaritriose a n d higher oligosaccharides. The enzyme is a type of e n d ofl-l,3-glucanase.
