Centrifugation
in density gradients is an important procedure for the fractionation and characterization of macromolecules and cellular organelles. A number of gradient-forming devices have been described (l-11), each with some desirable features. I now find that any one of several commercially available multichannel peristaltic pumps can be used to construct a simple and highly versatile gradient-forming system incorporating the following advantages: (1) rapid operation ; (.z?) capacity to form two (or more) identical gradients at a time; (3) great volume versatility, making it suitable for tubes and, potentially, for zonal rotors; (4) capacity to form linear, convex, and concave gradients. The basic principles of the system have been detailed by Wallach and Nordby (12) ; its application is shown here in the formation of small-volume gradients for membrane fractionation (13).
