✦ Synopsis
A simple method for microdetermination of muramic acid is elaborated. The method is based on the degradation of muramic acid to lactic acid, followed by degradation of the latter to acetaldehyde which can be determined calorimetrically with p-hydroxydiphenyl (PHD) A linear relationship exists between the concentration of muramic acid (up to 20 pg), and absorbance at 569 nm. Substances usually present in the hydrolysates of bacterial cell wall peptidoglycan do not interfere in the determination.

A new method for determination of uronic acids with meta-hydroxydiphenyl is introduced. It is simpler, quicker, more sensitive, and more specific than other methods, and it needs lesser amounts of fluid. It is recommended for determination of acid mucopolysaccharides in biological materials.
