A new technique
is described which allows determination of solute uptake into cultured cell monolayers at times approaching 0.5 s. The apparatus consists of a section of a plastic tissue culture multidish which is held in a rotatable clamp and a manifold for simultaneous washing of the multidishes with ice-cold buffer. This technique and apparatus was used to measure the initial rates of nucleoside and base transport by Chinese hamster fibroblasts.
