✦ Synopsis

Thin-layer chromatogram (6 x 6 cm) of the model mixture: (1) inorganic phosphate, (2) choline, (3) choline phosphate, (4) glycerophosphorylcholine, (5) glycerophosphate, (6) betaine. Solvent systems: First direction (vertical), methanol/water/conc. NH4 OH/chlorotorm/acetone (5 : 2 : 1 : 2 : 2); Second direction (horizontal) methanol/water/conc. NH40H (5 : 3 : 1).

Visualization was rendered with iodine vapours (the second and the sixth spots) and malachite green reagent consecutively.

We describe a quantitative assay for diaminopimelic acid (DAP) in urine. It involves (i) hydrolysis of urine samples, (ii) purification by several different liquid chromatography steps, and (iii) analysis by high-performance liquid chromatography on a reversed-phase C18 column. Dihydropyrimidine dehydrogenase (DPD) is the initial, rate-limiting enzyme in the catabolism of 5-fluorouracil (5-FU). A pharmacogenetic syndrome has been described in which DPD-deficient patients are at risk for toxicity following administration of 5-FU. To date, there are at least 21 previously de
