✦ Synopsis
Membrane preconcentration transient isotachophoresis capillary elec-Ž . Ž . trophoresis mPC-tITP-CE was coupled to an ion trap mass spectrometer MS Ž . through a porous glass joint ion electrospray ionization source -ESI source . This novel CErMS interface gave enhanced signal with less chemical background while proving to be both durable and easy to manufacture. The utility of the mPC-tITP-CEr-ESIrMS combination for providing a separation orthogonal to Ž . high-performance liquid chromatography HPLC with detection limits compatible with immunological samples was demonstrated using a melanoma tumor cell extract from the DM6 cell line. This system easily resolved the antigen peptide from other candidate peptides not differentiated after multiple dimensions of HPLC. Detection limits of -50 amol in full-scan MS and MSrMS modes were observed. This system was not concentration limited.
