✦ Synopsis

A micromethod for the determination of arylsulfatase activity in cultured fibroblasts or amniocytes is described. The method depends on the hydrolysis of methylumbelliferyl sulfate in an assay volume of 0.3 ~1 using extracts from as few as lo-20 cells. Arylsulfatase A and B activities are distinguished using the silver ion inhibition method of H. Christomanou and K. Sandhoff (1977) C/in. Chim. Acla 79, 527-531. The method is illustrated for the detection of metachromatic leukodystrophy resulting from a deficiency of arylsulfatase A activity.
