A highly sensitive assay for S-adenosylmethionine (AdoMet) by high-performance liquid chromatography with amperometric detection is described. The method is based on the measurement of 3-methoxytyramine formed from AdoMet and dopamine in the presence of catechol-O-methyltransferase. Using this technique 1 pmol of AdoMet can be measured and the AdoMet content of less than 1 mg of adrenal tissue can be accurately determined. This method is particularly suitable for assay of AdoMet in small amounts of tissues obtained by microsampling techniques.
