โฆ Synopsis
A spectrophotometric method for the analysis of the enzymatic hydrolysis of N-acyl peptides has been developed based on the reaction of amino acids with phthaldialdehyde in the presence of thiols. The method approaches the sensitivity of ninhydrin while being more simple and rapid. In addition, the reagent can be prepared, stored, and used without special handling. The substitution of ethanethiol for mercaptoethanol greatly enhances the stability of the colored product. The method has been applied to the study of the kinetics of the hydrolysis of N-acyl peptides by thermolysin and pepsin.
