✦ Synopsis

We described a chemiluminexent immunoassay (CIA) for 5a-androstane-3a,l7~-diolglucuronide (3a-diol-G) in human diluted urine. This method allowed the direct measurement in 1 pl of urine avoiding the hydrolysis and extraction steps for sample pretreatment commonly used in routine methods. The hapten 3a-diol-G was synthesized by a Koenigb-Knorr reaction. The immunogenic complex, 3a-diol-G conjugated t o bovine serum albumin (BSA), was employed t o induce the formation of specific antibodies in New Zealand rabbits. In addition, the required chemiluminescent (CL) tracer was prepared. The characteristics of the antibody was determined as regard to specificity and sensitivity and the precision of the assay methods established. In 22 hirsute women affected by policystic ovarian syndrome we found 3a-diol-G values significantly (p -= 0.01) higher (146.28 k 73.77pgIg of creatinine; mean f SD) than those observed in normal women (72.1 k 32.58pg/g of creatinine; mean zt SD).

Pregnanetriol-3a-glucuronide (PTG) is the majority urinary metabolite of 17hydroxyprogesterone (170HP) and it typically increases in the commonest form of congenital adrenal hyperplasia (CAH), due t o 21 hydroxylase deficiency. We developed a simple chemiluminescent immunoassay for the direct measu
