Abstract

The synthesis of [^14^C] labelled Sulodexide is reported. Sulodexide is a sulphated polysaccharide of the class of glycosaminoglycan, containing a heparin like fraction (70%), dermatan sulphate (20%) and other minor fractions. The heparinโ€like fraction, suitably isolated from other components, was partially and selectively Nโ€desulphated, thus making few percent unit โ€NH~2~ groups available for the labelling with [^14^C]โ€acetic anhydride (specific activity 0.15 ฮผCi/mg). Due to the small extent of modification, the sulphate to carboxylate group ratio remained practically unchanged on the heparinโ€like fraction. Sulodexide was reconstituted adding to the labelled fraction the suitable amount of the other components; the chemical and biological properties of the final labelled Sulodexide were undistinguishable from those of the starting material.

Procedures for the preparation of UDP-N-[1-14C]acetyl-D-glucosamine and UDP-N-[1-14C]acetyl-D-galactosamine with very high specific activities are described. The overall yield based on the amount of [1-14C]acetate used is greater than 80%. The N-acetyl-D-glucosamine-alpha-1-phosphate used in this sy
